How do I run a DOS app. in Win32?

I am trying to run a DOS game in dosemu and DosBox but when I try to activate said DOS program, it says it must be run under Win2.
I have WINE installed, but I also don't know how to run a DOS app on it. What package do I need to install to get my DOS app to work and play?
